What you are looking for is actually the function in your Internet Browser for automatically opening downloaded files, rather than an EndNote setting. This will trigger the file to automatically open when downloaded, providing a seamless process.
Truth be told, it isn’t possible for EndNote to import a file that hasn’t been downloaded to your system.
But, setting that file to open as soon as the download is finished should help streamline importing.
